Principle of clean-up

Mycotoxins occur in various commodities, many of them are highly complex. Compared to the amounts of other components in agricultural commodities like carbohydrates, fatty acids or proteins, the amounts of mycotoxins present in a sample are incredibly low. Our clean-up columns must fulfill certain criteria:
  • Separate the toxin of interest from matrix interferences
  • Recover the toxin of interest quantitatively
  • Save time and costs

Our range of clean-up columns (single step multifunctional or immunoaffinity columns -IACs) for sample preparation prior to TLC, HPLC, GC, mass spectroscopy and fluorometry provides you with the most complete choice available. Star™ Immunoaffinity Columns (IACs)

StarTM immunoaffinity columns are reliable and user-friendly, designed for rapid and simple purification of mycotoxins from complex matrices, in cases where extremely low limits of detection are required (e.g. determination of toxins in baby food).

Working Principle
Immunoaffinity columns contain antibodies that selectively bind the toxin of interest from the crude extract. During the clean-up step, interferences can be washed from the column while the toxin is immobilized on the column. In the last step, the toxin is eluted from the column for determination purposes. Using this highly specific technology, almost all clean-up related issues can be solved.

MycoSep® Columns

MycoSep® columns remove analytical interferences from sample extracts in one quick, easy step, eliminating washing and elution steps found in solid phase extraction and affinity columns. This patented system allows multifunctional clean-up for HPLC, mass spectroscopy (MS), GC, fluorometry and TLC analysis of several different mycotoxins, with an optimal precision and accuracy.

Working Principle

MycoSep® columns allow a one-step clean-up within 30 seconds: The MycoSep® clean-up column is pushed into a test tube (containing the sample extract), forcing the extract to filter upwards through the packing material of the column. The interferences adhere to the chemical packing in the column and the purified extract, containing the analytes of interest, passes through a membrane (frit) to the surface of the column.

Why use MycoSep® clean-up?

  • If the alternative is liquid-liquid partition, the lower consumption of hazardous solvents and the huge reduction of working time are clear advantages of MycoSep® columns. The simple one step clean-up with MycoSep® excludes many sources of error.
  • If the alternative method is an immunoaffinity column specific only for deoxynivalenol, the MycoSep® clean-up is superior because it also allows the analysis of other type B trichothecenes, like Nivalenol, Fusarenon X, 3- and 15-Acetyldeoxynivalenol and type A trichothecenes, which is not possible after immunoaffinity clean-up. This method of analysis gives a more complete picture of the contamination of the sample with trichothecenes.
  • If the alternative is an immunoaffinity column, MycoSep® clean-up columns have the additional advantage of speed: clean-up takes only 30 seconds compared to approximately 30 minutes when clean-up is done with immunoaffinity columns.

MultiSep® Columns

MycoSep® clean-up columns are also available in our MultiSep® syringe format, giving our customers higher flexibility in choosing their preferred format. MultiSep® columns are not only used for direct sample clean-up before HPLC, Mass spectrometry (MS), GC, etc. and they can also be used when additional clean-up is necessary.

Working Principle
Similar to the MycoSep® column format, the MultiSep® column format allows a one-step clean-up of sample extracts, containing the analyte of interest, within 30 seconds: The sample extract is applied on top of the MultiSep® clean-up column, allowing the extract to go through the packing material of the column by gravity. Alternatively, MultiSep® columns can be attached to SPE stations, thus allowing the application of vacuum or positive pressure to force the extract through the column material. The interferences adhere to the chemical packing in the column and the purified extract, containing the analytes of interest, can be easily be collected in a suitable vessel.

 

Why use MultiSep® clean-up?

  • If the alternative is liquid-liquid partition, the lower consumption of hazardous solvents and the huge reduction of working time are clear advantages of MultiSep® columns. The simple one-step clean-up procedure with MultiSep® columns excludes many sources of error.
  • If the alternative is an immunoaffinity column, MultiSep® clean-up columns have the additional advantage of speed: clean-up takes only 30 seconds compared to approximately 30 minutes when clean-up is done with immunoaffinity columns.
  • The MultiSep® column format in combination with SPE stations offers the opportunity to purify several different samples simultaneously.
